FindingNemo Toolkit Enhances Ultra-Long DNA Sequencing in a Single Day

A new toolkit named FindingNemo has been developed for DNA extraction, library preparation, and purification for ultra-long nanopore sequencing. The protocol, which is a result of a collaborative effort involving researchers from institutions such as DeepSeqNotts, can be adapted to work with as few as one million human cell equivalents or 5 micrograms of ultra-high molecular weight DNA. It allows for extraction to sequencing in a single working day. The toolkit is designed to produce high occupancy ultra-long reads on nanopore platforms, offering throughput comparable to disc-based methods and potential advantages for tissues and non-human cell materials. This development also aids in understanding allele-specific expression in single-nucleus RNA-seq data.
